Stability Studies — ICH Q1A(R2), Zone IVb

Long-term, intermediate, accelerated and photostability studies run in mapped chambers with calibrated monitoring, pre-approved protocols, defined pull schedules and out-of-trend results investigated rather than averaged away.

Storage conditions we run

Why Zone IVb is the condition that matters here

India, the Gulf and much of South East Asia sit in climatic Zone IVb, so long-term storage is 30 °C / 75% RH rather than the 25 °C / 60% RH used in temperate markets. Filing Zone II data for a Zone IVb market is a predictable deficiency, and a product that holds at 25 °C does not automatically hold five degrees and fifteen points of humidity higher.

Pull schedule and study design

Long-term pulls at 0, 3, 6, 9, 12, 18, 24 and 36 months; accelerated at 0, 3 and 6; intermediate triggered when a significant change appears under accelerated conditions. Bracketing and matrixing designs are used where strengths or container sizes justify them, with the justification written into the protocol rather than applied afterwards.

What gets tested

Assay and related substances by HPLC or UPLC, dissolution for oral solids, water content by Karl Fischer for lyophilised products, pH, osmolality, colour and clarity, sub-visible particulate matter, container closure integrity, preservative content and microbial limits or sterility as applicable. For cosmetics: viscosity, globule size, centrifugation, freeze-thaw cycling and preservative efficacy to ISO 11930.

Out-of-trend handling

A result inside specification can still predict failure. A related substance moving from 0.08% to 0.31% against a 0.5% limit passes every pull and will not survive the claimed shelf life. Each result is evaluated against the established trend as well as the limit, and an out-of-trend result triggers the same investigation rigour as an out-of-specification one.

Chamber qualification

Chambers are temperature and humidity mapped at installation and on requalification, loaded and empty, with calibrated independent sensors. Monitoring is continuous with alarm escalation and a documented excursion procedure, because a study is only as defensible as the evidence that the chamber held condition for its full duration.

Starting a study

Send the formulation, container closure system, specification and the markets you are filing in to Stancor Pharma. We will propose a protocol with conditions, pull schedule, test panel and the shelf life the data set can support — including whether extrapolation is defensible or a committed stability study is needed.
